Explain why spinal segment #25 does not lie under the vertebra #25(lumbar vertebra #5, or L5)...?

1. Why is this knowledge useful in performing a spinal tap(lumbar puncture)?
This is only a guess, but there are 3 different areas of your spinal vertebrae: lumbar (lower), thoracic (lower-upper back) and cervical neck to the base of skull. I think they each have their own sets of numbers and maybe this is why. As for the lumbar puncture, I have had two and it makes a difference to the doctor performing the procedure that they get between two vertebrae with a wide enough opening to perform it safely. That's why they have you bent in as tight of a fetal position as possible to do the procedure because it widens those spinal spaces.
